Pharmacology and Toxicology: Proton pump inhibitors. This product is a lipid-soluble, weakly basic drug that readily accumulates in acidic environments. After oral administration, it specifically distributes into the secretory canaliculi of gastric parietal cells, where it transforms into its active sulfonamide form under highly acidic conditions. This active form then forms an irreversible covalent bond with the sulfhydryl groups of the H⁺,K⁺-ATPase enzyme (also known as the proton pump) located in the parietal cell's secretory membrane—specifically through a disulfide bridge. The resulting sulfonamide-proton pump complex effectively inhibits the enzyme's activity, blocking the final step of gastric acid secretion. As a result, this medication exerts a potent and long-lasting suppression of acid production, regardless of the underlying cause.
